| 背景信息 | ORC ubiquitin ligase 1 (OBI1) is a human E3 ubiquitin ligase essential for the activation of DNA replication origins during the S phase of the cell cycle. It functions as a replication origin selector by localizing to chromatin and catalyzing the multi-mono-ubiquitination of specific subunits within the Origin Recognition Complex (ORC), namely ORC3 and ORC5. This post-translational modification is crucial for the selection and firing of replication origins, likely enhancing the assembly of the pre-replication complex. OBI1 exhibits chromatin binding and ubiquitin-protein transferase activities, facilitating protein autoubiquitination and monoubiquitination to regulate the fidelity of DNA replication. By complementing the ATPase activities of the ORC complex, OBI1 ensures the precise temporal and spatial control of genomic duplication. |
