| 背景信息 | E4F transcription factor 1 (E4F1) is a zinc finger protein of the GLI-Kruppel family that serves as both a transcriptional repressor and an E3 ubiquitin ligase. It plays a central role in regulating cell cycle progression, DNA damage response, and metabolism by mediating the ubiquitination of chromatin-associated TP53 (p53). E4F1 also regulates the transcription of cyclin CCNA2 and interacts with key tumor suppressors such as p16 and RB1. Its activity is modulated by adenovirus E1A through proteolytic cleavage and phosphorylation, which can convert it into a transcriptional activator. Furthermore, E4F1 cooperates with PARP-1 and BRG1 to facilitate DNA double-strand break repair, highlighting its importance in maintaining genomic stability and cellular survival. |
