| 背景信息 | STEAP4 metalloreductase (STEAP4) is an integral membrane protein localized to the Golgi apparatus and mitochondria that functions as a NADPH-dependent metalloreductase. It facilitates the reduction of Fe(3+) to Fe(2+) and Cu(2+) to Cu(1+), playing a vital role in cellular iron and copper homeostasis, particularly within adipocytes, hepatocytes, and pancreatic islets. Structurally, STEAP4 operates as a homodimer containing an N-terminal oxidoreductase domain and a C-terminal six-helix transmembrane domain. It serves as a critical link between inflammatory and metabolic responses, with expression induced by TNF-alpha. Clinically, genetic variants of STEAP4 are associated with obesity and insulin resistance, while its dysregulation is linked to mitochondrial iron overload, inflammatory arthritis, and the progression of colitis-associated colon cancer. |
