| 背景信息 | Ring finger protein 43 (RNF43) is a transmembrane E3 ubiquitin ligase that serves as a critical negative regulator of the Wnt signaling pathway. It contains a RING finger domain and an ectodomain, through which it mediates the ubiquitination and subsequent endocytosis of Frizzled receptors, thereby reducing their surface expression and suppressing Wnt activation. RNF43 functions as a significant tumor suppressor; mutations in this gene lead to aberrant beta-catenin signaling and are frequently observed in various malignancies. Clinically, N-terminal mutations in RNF43 can sensitize cancer cells to Wnt antagonists, and its status serves as a prognostic indicator in clear cell renal cell carcinoma. Additionally, RNF43 mutations may predict therapeutic response to anti-BRAF/EGFR combinations, highlighting its utility as both a biomarker and a therapeutic target. |
