| 背景信息 | Fc receptor like 6 (FCRL6) is a type I transmembrane immunoregulatory glycoprotein predominantly expressed on mature cytotoxic CD8+ T cells and NK cells. Unlike classical Fc receptors, FCRL6 functions as a receptor for MHC class II molecules, specifically binding HLA-DR. Its cytoplasmic tail contains tyrosine-based motifs, including an ITIM-like sequence that recruits SHP-2 phosphatase to mediate inhibitory signaling in cytotoxic lymphocytes. FCRL6 expression increases with lymphocyte differentiation and cytolytic potential, suggesting a role in modulating immune responses. In the context of oncology, FCRL6 is upregulated on lymphocytes infiltrating HLA class II-expressing tumors, where it contributes to immune tolerance and tumor evasion. It is also associated with combined immunodeficiency and thrombocytopenia in genetic studies. |
