| 背景信息 | Coiled-coil domain containing 137 (CCDC137) is a versatile nucleolar protein that modulates ribosome biogenesis, nuclear receptor signaling, and chromatin-related processes. It functions as an RNA-binding protein that enhances the cytoplasmic localization and expression of various mRNAs, such as FOXM1 and LASP1, thereby activating AKT signaling to drive cell proliferation and the G1/S transition. CCDC137 also acts as a sensor of nucleolar stress, particularly during HIV-1 infection, where it responds to Vpr-induced DNA damage. Clinically, CCDC137 is overexpressed in several malignancies, including breast, kidney, and lung cancers, where its levels correlate with immunosuppressive markers and poor prognosis. Additionally, mutations in CCDC137 have been associated with familial thyroid dyshormonogenesis. |
