| 背景信息 | RPTOR independent companion of MTOR complex 2 (RICTOR) is a protein that forms part of the mammalian target of rapamycin complex 2 (mTORC2). It plays a crucial role in integrating nutrient- and growth factor-derived signals to regulate cell growth and proliferation. Structurally, RICTOR contains few conserved regions and has 21 identified phosphorylation sites, with T1135 undergoing growth factor-responsive phosphorylation via S6K1. Functionally, RICTOR is essential for the phosphorylation of Akt/protein kinase B (PKB) on SER473, which activates Akt/PKB and is implicated in cancer and diabetes. RICTOR has been associated with various cancers, including colorectal carcinoma, glioblastoma, and pituitary adenomas, where its overexpression can lead to increased cell proliferation and survival. In embryonic development, RICTOR and mTORC2 are necessary for actin cytoskeleton organization and embryonic growth. |
