| 背景信息 | Layilin (LAYN) is a single-pass type I transmembrane protein characterized by an extracellular C-type lectin domain that functions as a specific receptor for hyaluronan. It mediates cell adhesion, motility, and cortical organization by interacting intracellularly with cytoskeletal adaptors such as talin, merlin, and radixin. These interactions link the plasma membrane to the actin cytoskeleton, particularly at membrane ruffles during cell spreading and migration. Layilin is widely expressed across various tissues and has gained prominence in tumor immunology. In the tumor microenvironment, it is highly expressed on exhausted CD8+ T cells, where it augments LFA-1 integrin activation and modulates T cell cytotoxicity. Research indicates that layilin plays a significant role in regulating antitumor immunity, as its presence or deletion directly impacts T cell function and tumor growth in clinical models. |
