| 背景信息 | Unc-5 netrin receptor D (UNC5D) is a type I transmembrane receptor that primarily binds netrin-4 to regulate axon guidance, neuronal migration, and cell-cell adhesion. In the developing nervous system, UNC5D mediates the repulsion of neuronal growth cones and directs the radial migration of cortical neurons. Functioning as a dependence receptor, it promotes cell survival when engaged by its ligand; however, in the absence of netrin or following DNA damage, the receptor is cleaved by caspases to trigger apoptosis via its intracellular death domain. This dual role makes UNC5D a critical regulator of programmed cell death. Clinically, UNC5D expression is a significant prognostic marker in neuroblastoma, where its involvement in a p53/E2F1-linked pro-apoptotic loop is associated with favorable clinical outcomes. |
