| 背景信息 | Autophagy related 14 (ATG14), also known as BAKOR, is an essential regulator of autophagy that functions as a core component of the class III phosphatidylinositol 3-kinase (PI3KC3-C1) complex. It directs the localization of this complex to initiate autophagosome formation and facilitates MAP1LC3 conjugation and BECN1 translocation. ATG14 also promotes autophagosome-endolysosome fusion by interacting with STX17-SNAP29 binary t-SNARE complexes. Beyond its role in autophagy, ATG14 is critical for maintaining mitochondrial integrity and preventing pyroptosis. Clinically, loss of ATG14 has been linked to severe structural abnormalities in the oviduct and infertility, and it is also associated with neurodegeneration with brain iron accumulation. |
