| 背景信息 | Ring finger protein 207 (RNF207) is a ubiquitin-protein ligase characterized by an N-terminal RING domain that plays a critical role in cardiac electrophysiology. It regulates action potential duration by interacting with the KCNH2/HERG potassium channel, enhancing its trafficking, membrane expression, and current density. This regulatory mechanism is dependent on heat shock protein 70 (HSP70), which binds to RNF207 to facilitate channel stability. Functional studies indicate that RNF207 deficiency prolongs action potentials and can lead to atrioventricular block, while its overexpression shortens them. Clinically, genetic variants in RNF207 are well-established risk factors for prolonged QT interval, which predisposes individuals to ventricular arrhythmias such as torsades de pointes and sudden cardiac death. |
