| 背景信息 | TAO kinase 1 (TAOK1) is a STE20-family serine/threonine-protein kinase that associates with the microtubule cytoskeleton and nuclear bodies. It serves as an upstream activator of the p38/MAPK14 stress-activated cascade by phosphorylating MAP2K3 and MAP2K6. TAOK1 also regulates cytoskeletal stability by activating MARK2, which promotes the detachment of tau from microtubules, and is essential for proper chromosome congression and mitotic progression. Clinically, heterozygous pathogenic variants in TAOK1 cause an autosomal dominant neurodevelopmental disorder characterized by developmental delay and intellectual impairment. Additionally, TAOK1 is implicated in the pathogenesis of Alzheimer-related tau pathology and has been linked to inflammatory IL-17 signaling and fibrogenesis, suggesting a broad role in both developmental and degenerative diseases. |
