| 背景信息 | (Trpm4) is a calcium-activated monovalent cation channel that forms homotetrameric structures to regulate cellular membrane potential. Each subunit features six transmembrane helices and an extensive N-terminal domain that facilitates interactions with intracellular signaling molecules. TRPM4 conducts sodium and potassium ions without calcium permeability, thereby depolarizing cells to modulate action potential shape, insulin secretion, and immune cell migration. Its activity is finely tuned by calcium binding, ATP inhibition, and post-translational modifications such as SUMOylation. Clinically, gain-of-function mutations in TRPM4 are a well-established cause of progressive familial heart block type I, while altered expression levels are linked to cardiac hypertrophy and various cancers. |
