| 背景信息 | HDGF like 2 (HDGFL2) is a hepatoma-derived growth factor-like protein that functions as a histone reader through its N-terminal PWWP domain. It binds various methylated histones, including H3K36me3 and H4K20me2/3, to facilitate epigenetic regulation and chromatin remodeling. HDGFL2 plays a vital role in myogenesis by cooperating with the BAF complex and contributes to DNA double-strand break repair via the recruitment of RBBP8. Furthermore, it supports cellular growth and influences viral integration sites during HIV-1 replication. In clinical contexts, HDGFL2 is highly relevant to neurodegenerative proteinopathies; cryptic exon inclusion resulting from TDP-43 dysfunction produces stable HDGFL2 variants that serve as specific biomarkers in the cerebrospinal fluid of patients with frontotemporal dementia and amyotrophic lateral sclerosis. |
