| 背景信息 | WAPL cohesin release factor (WAPL) is a fundamental regulator of the cohesin complex, essential for maintaining sister chromatid cohesion and three-dimensional genome architecture. It promotes the unloading of cohesin from chromatin by interacting with Pds5, driving a dynamic cycle of loading and release that influences cell-type-specific gene expression. During meiosis, WAPL ensures proper chromosome segregation by selectively releasing cohesin from chromosome arms. It also interacts with Bub3 to support spindle assembly checkpoint activity. Dysregulation of WAPL disrupts cohesin dynamics and is associated with cohesinopathies, most notably Cornelia de Lange syndrome, where altered chromatin looping leads to significant developmental gene dysregulation. |
