| 背景信息 | Desmoglein 4 (DSG4) is a calcium-dependent cadherin and a core transmembrane component of desmosomes, essential for cell-cell adhesion in epithelial tissues. It is highly expressed in hair follicle keratinocytes and the upper layers of the epidermis, where it coordinates the transition from cellular proliferation to differentiation. DSG4-mediated desmosomes provide mechanical strength and regulate signaling pathways necessary for hair shaft formation and epidermal barrier integrity. Clinically, mutations in DSG4 are the primary cause of localized autosomal recessive hypotrichosis (Hypotrichosis 6), characterized by fragile, sparse hair due to underdeveloped hair follicles. DSG4 variants are also associated with monilethrix-like congenital hypotrichosis, a disorder of the hair cortex resulting in beaded hair shafts and significant hair dysplasia. |
