| 背景信息 | RPA interacting protein (RPAIN) is a nuclear chaperone that mediates the nuclear import of the heterotrimeric Replication Protein A (RPA) complex. By acting as a functional scaffold, RPAIN stabilizes the extended conformation of RPA’s multiple DNA-binding domains, preventing premature or non-specific interactions with other proteins in the absence of single-stranded DNA (ssDNA). Upon DNA availability, it promotes the formation of high-density RPA-ssDNA filaments essential for replication and repair. RPAIN likely utilizes interactions with importin beta for nucleocytoplasmic shuttling and may be modulated by SUMOylation. While RPA dysregulation is linked to replication stress, direct clinical associations for RPAIN itself remain unestablished in primary human disease contexts. |
