| 背景信息 | Zinc finger protein 598, E3 ubiquitin ligase (ZNF598) is a critical component of the ribosome quality control (RQC) pathway, where it functions as a sensor for stalled or collided ribosomes. Upon detecting ribosomal collisions, ZNF598 mediates the monoubiquitination of 40S ribosomal proteins RPS10 and RPS3, as well as the Lys-63-linked polyubiquitination of RPS20 via the E2 enzyme UBE2D3. These modifications trigger the recruitment of the RQT complex for ribosome disassembly and the degradation of nascent peptides. Additionally, ZNF598 recruits the 4EHP-GYF2 complex to repress translation during development and supports viral protein synthesis. Clinically, genetic variants in ZNF598 are associated with frontotemporal dementia and amyotrophic lateral sclerosis 7 (FTD/ALS7), where impaired protein homeostasis contributes to neurodegeneration. |
