| 背景信息 | Family with sequence similarity 83 member A (FAM83A) is a cytoplasmic and nuclear protein characterized by a conserved DUF1669 domain. It functions as a potent regulator of the EGFR/MAPK and PI3K/AKT signaling pathways and is involved in mitochondrial maintenance. FAM83A directly interacts with beta-catenin and is phosphorylated by SRC-family kinases, which stabilizes beta-catenin and promotes its nuclear translocation to enhance Wnt target gene transcription. Pathologically, FAM83A acts as a transforming oncogene and is frequently overexpressed in several malignancies, most notably breast and pancreatic cancers. Its overexpression drives tumor proliferation, invasion, and anchorage-independent growth, while also conferring resistance to EGFR and PI3K inhibitors. Consequently, FAM83A is recognized as a significant therapeutic target for overcoming drug resistance in cancer treatment. |
