| 背景信息 | Homer scaffold protein 1 (HOMER1) is a critical postsynaptic density protein that organizes synaptic signaling complexes. It exists as long-form tetramers (Homer1b/c) that cross-link group 1 metabotropic glutamate receptors (mGluR1/5), IP3 receptors, and SHANK proteins, thereby coupling surface receptors to intracellular calcium stores. An activity-dependent short-form variant, Homer1a, acts as an immediate early gene that disrupts these scaffolds to facilitate synaptic plasticity and dendritic spine remodeling. HOMER1 is widely expressed in the central nervous system and is essential for memory formation and neuronal homeostasis. Clinically, HOMER1 dysfunction is well-documented in Alzheimer’s disease and other neurological disorders, where disrupted expression levels impair synaptic transmission and contribute to cognitive decline. |
