| 背景信息 | Ring finger protein 135 (RNF135) is an E3 ubiquitin-protein ligase that plays a pivotal role in antiviral innate immunity. It contains a RING finger domain that mediates Lys-63-linked polyubiquitination and serves as a coreceptor for RIG-I (DDX58). Upon sensing viral RNAs in the cytoplasm, RNF135 ubiquitinates RIG-I oligomers and synthesizes unanchored polyubiquitin chains to enhance immune signaling. Beyond its immunological role, RNF135 is involved in oncogenic pathways, promoting proliferation and metastasis in triple-negative breast cancer and glioblastoma via ERK signaling. Clinically, the RNF135 gene is located within the 17q11.2 region frequently deleted in neurofibromatosis type 1 (NF1), and its loss is associated with NF1 microdeletion syndrome and an increased risk of malignancy. It has also been linked to Autism Spectrum Disorder. |
