| 背景信息 | BR serine/threonine kinase 2 (BRSK2) is a serine/threonine-protein kinase essential for neuronal polarization, axonogenesis, and the regulation of mitotic cell cycle progression. Activated by the upstream kinase STK11/LKB1, BRSK2 localizes to centrosomes and the endoplasmic reticulum, where it phosphorylates microtubule-associated proteins such as MAPT/TAU to modulate cytoskeletal dynamics. Beyond its neurodevelopmental roles, it regulates insulin secretion via PAK1-mediated glucose responses and participates in ER stress-induced apoptosis. The protein is highly expressed in the brain and lacks signal peptides or transmembrane regions. Mutations in the BRSK2 gene are established causes of Autosomal Dominant Non-Syndromic Intellectual Disability, reflecting its critical role in neurodevelopment and synaptic function. |
