| 背景信息 | Hyccin PI4KA lipid kinase complex subunit 2 (HYCC2), also known as FAM126B, is a regulatory subunit of the plasma membrane phosphatidylinositol 4-kinase IIIα (PI4KA) complex. It forms a constitutive core with PI4KA and TTC7, assembling into a large supercomplex responsible for generating phosphatidylinositol 4-phosphate (PI4P). This lipid is a critical precursor for PI(4,5)P2 and other phosphoinositides essential for plasma membrane signaling and homeostasis. HYCC2 provides a broad membrane-interacting surface that stabilizes PI4KA and orients its catalytic domain toward the lipid bilayer for efficient synthesis. Clinically, the integrity of the PI4KA-TTC7-FAM126 complex is vital for myelinating glia and membrane biogenesis. Disruption of this complex is associated with severe developmental disorders, including hypomyelination, congenital cataracts, and combined immunodeficiency, underscoring its fundamental role in neurological and immunological health. |
