| 背景信息 | Structural maintenance of chromosomes 5 (SMC5) is a core component of the SMC5-SMC6 heterodimeric complex, which is essential for maintaining genomic integrity. This ATP-binding protein plays a critical role in DNA double-strand break repair via homologous recombination, sister chromatid cohesion during mitosis, and telomere maintenance. SMC5 localizes to the nucleus and chromosomes, where it interacts with cohesin and shelterin complexes to facilitate recombination and suppress the transcription of episomal DNA. In meiosis, the complex ensures genomic stability by promoting homolog-independent sister-chromatid recombination. Clinically, mutations in SMC5 are associated with Atelis Syndrome 2, a neurodevelopmental disorder characterized by primordial dwarfism and microcephaly. |
