| 背景信息 | SCY1 like pseudokinase 3 (SCYL3), also known as PACE-1, is a member of the SCYL protein family characterized by an inactive kinase domain, central HEAT repeats, and a disorganized C-terminal segment. Unlike conventional kinases, SCYL3's pseudokinase domain lacks catalytic activity but may play roles in protein trafficking and signaling pathway assembly. Unique to SCYL3 is a myristoylation consensus sequence at the N-terminus, which facilitates membrane association. SCYL3 interacts with ezrin through its C-terminal domain and has been implicated in intracellular processes such as clathrin-mediated endocytosis and neuronal function. Clinically, SCYL3 is overexpressed in hepatocellular carcinoma (HCC), where it promotes tumor progression by regulating ROCK2 activity. Patients with high SCYL3 expression exhibit reduced survival rates. Additionally, SCYL3 plays overlapping roles with SCYL1 in motor neuron viability, and its disruption accelerates neurodegenerative conditions resembling amyotrophic lateral sclerosis (ALS). |
