| 背景信息 | Adhesion G protein-coupled receptor F4 (ADGRF4), also known as GPR115, is a class B2 adhesion GPCR characterized by a large extracellular region and a GAIN domain. It typically undergoes autoproteolytic cleavage to form noncovalently associated fragments that regulate receptor activation and transmembrane signaling. ADGRF4 is predominantly expressed in the epidermis, specifically within spinous and granular keratinocytes, where it functions as a key regulator of epidermal differentiation and KRT1 expression. This role is essential for proper keratinocyte maturation and stratification. Additionally, ADGRF4 is involved in the development of other ectodermal tissues, such as enamel mineralization by ameloblasts. Clinically, mutations in ADGRF4 have been linked to complex cortical dysplasia and other brain malformations, highlighting its broader significance in tissue development and morphogenesis. |
