| 背景信息 | Calcium/calmodulin dependent protein kinase kinase 1 (CAMKK1) is a serine/threonine kinase belonging to the Ca2+/calmodulin-dependent protein kinase (CAMK) family, characterized by a kinase domain with an N-terminal lobe of β-strands and a predominantly α-helical C-terminal lobe. It contains an auto-inhibitory domain overlapping with a calmodulin-binding domain at the C-terminus, which keeps the enzyme inactive until calcium/calmodulin binding activates it. CAMKK1 functions as an upstream kinase in the calcium-triggered signaling cascade, phosphorylating and activating downstream kinases such as CAMK1 and CAMK4, thereby regulating transcription factors, cell cycle, hormone production, neurite outgrowth, and actin cytoskeleton organization. Structural studies have revealed its ATP-binding pocket and interactions with potent inhibitors, aiding selective drug design. Dysregulation of CAMKK1 is implicated in neurological disorders due to its role in neuronal maturation, axon elongation, and long-term memory formation. Additionally, CAMKK1 is involved in cardiovascular signaling pathways and may contribute to disease states related to calcium signaling abnormalities. |
