| 背景信息 | Ring finger protein 169 (RNF169) is an E3 ubiquitin-protein ligase that acts as a critical negative regulator of the ubiquitin-dependent DNA damage response (DDR). It localizes to double-strand breaks (DSBs) in an RNF8/RNF168-dependent manner, utilizing its MIU domains to compete for ubiquitin structures at the damage site. By antagonizing RNF168-mediated ubiquitylation, RNF169 attenuates the accumulation of downstream signaling proteins such as 53BP1 and RAP80. This inhibitory mechanism promotes high-fidelity homologous recombination repair over error-prone non-homologous end joining, thereby maintaining genomic stability. The stability and recruitment of RNF169 are further regulated by the deubiquitinase USP7. Its primary function in DSB signaling is driven by its ability to bind ubiquitin and modulate the recruitment of repair factors to chromatin. |
