| 背景信息 | Exophilin 5 (EXPH5) is a Rab27 effector protein that plays a critical role in intracellular vesicle trafficking and exosome secretion. It is particularly essential in keratinocytes, where it facilitates the delivery of lamellar bodies to the plasma membrane, a process vital for epidermal lipid extrusion and barrier formation. Structurally related to synaptotagmin-like proteins, EXPH5 also supports calcium-induced lysosomal exocytosis and peripheral lysosome trafficking. Loss of EXPH5 disrupts keratin filament organization and impairs the expression of differentiation markers such as filaggrin and keratin-10. Clinically, germline mutations in the EXPH5 gene result in skin fragility and a poorly differentiated epidermis, highlighting its necessity for maintaining epidermal homeostasis and structural integrity. |
