| 背景信息 | Semaphorin 6D (SEMA6D) is a type I transmembrane glycoprotein belonging to the class 6 semaphorin family, distinguished by a cysteine-rich sema domain essential for its functional specificity. This protein plays a critical role as an axon guidance molecule, showing growth cone-collapsing activity on dorsal root ganglion neurons and acting as a stop signal in target areas. SEMA6D preferentially binds to class A plexin receptors, especially Plexin-A1, guiding neuronal development, influencing cardiac morphogenesis, and mediating immune cell interactions such as those between dendritic cells and T cells. Its expression is highest in brain, kidney, placenta, and heart, with multiple isoforms regulated via alternative splicing. Beyond nervous system guidance, SEMA6D contributes to immune regulation by influencing late-phase T cell activity. Clinically, altered SEMA6D expression has been associated with several diseases, including gastric cancer, where it promotes tumor angiogenesis and progression, and shows links to autism spectrum disorder and inflammatory bowel disease. |
