| 背景信息 | Leukocyte immunoglobulin-like receptor subfamily B member 4 (LILRB4) is an inhibitory receptor predominantly expressed on myeloid cells, including monocytes, macrophages, and dendritic cells. This receptor plays a critical role in regulating immune responses by modulating the activation and function of these cells. LILRB4 contains two extracellular immunoglobulin-like domains and is characterized by immunoreceptor tyrosine-based inhibitory motifs (ITIMs) in its cytoplasmic tail, which facilitate the recruitment of phosphatases that negatively regulate signaling pathways. Dysregulation of LILRB4 has been implicated in several diseases, particularly in cancer and autoimmune disorders. In cancer, high expression levels of LILRB4 on tumor-associated macrophages can create an immunosuppressive microenvironment, aiding tumor progression and evasion of immune surveillance. Additionally, LILRB4 has been associated with systemic lupus erythematosus (SLE) and other inflammatory conditions, where its inhibitory effects may contribute to the dysregulation of immune responses. |
