| 背景信息 | DnaJ heat shock protein family (Hsp40) member B8 (DNAJB8) is a specialized chaperone that functions to suppress the aggregation and toxicity of misfolded proteins. It is particularly effective against polyglutamine-expanded proteins and tau seeds, utilizing its C-terminal serine-rich region and aromatic-rich motifs to facilitate oligomerization and substrate binding. Unlike many Hsp40 members, DNAJB8 can inhibit protein aggregation independently of canonical Hsp70 recruitment. In addition to its role in proteostasis, DNAJB8 is recognized as a cancer-testis antigen. In malignancies such as renal cell carcinoma, it promotes the maintenance of cancer stem-like cells and chemoresistance by inducing HSF1 and stabilizing TP53. Its restricted expression and high immunogenicity make it a significant target for cancer immunotherapy. |
