| 背景信息 | Ring finger protein 139 (RNF139), also known as TRC8, is a multi-pass transmembrane E3 ubiquitin ligase resident in the endoplasmic reticulum. It contains a RING-H2 finger domain and plays a central role in protein quality control and lipid homeostasis. RNF139 mediates the sterol-accelerated degradation of HMGCR and ubiquitinates other targets such as INSIG1, INSIG2, and MHC class I molecules. It also inhibits SREBP processing, thereby regulating cholesterol and fatty acid metabolism. Through its interactions with VHL and UBE2G2, RNF139 acts as a regulator of cell cycle progression, specifically inducing G2/M arrest. Clinically, RNF139 is a recognized tumor suppressor; germline t(3;8) translocations that inactivate the gene are associated with hereditary renal cell carcinoma and non-medullary thyroid cancer, highlighting its critical role in preventing renal oncogenesis. |
