| 背景信息 | LEO1 component of Paf1/RNA polymerase II complex (LEO1) is a highly conserved subunit of the PAF1 complex (PAF1C) that associates with RNA polymerase II throughout transcription elongation. It plays a critical role in regulating histone modifications, such as H2B ubiquitination, as well as chromatin remodeling and polyadenylation. Structurally, LEO1 forms a tight heterodimer with Paf1, which relies on Ctr9 as a scaffold to manage the RNA polymerase II pool during the DNA damage response. The Paf1/LEO1 heterodimer interacts with the C-terminus of Ela1 to recruit the Elongin-Cullin E3 ligase, facilitating Rpb1 ubiquitination and proteasomal degradation following DNA damage. Beyond its role in transcription, LEO1 is essential for maintaining cellular quiescence, controlling H3K9 methylation, and supporting embryonic stem cell maintenance across species. |
