| 背景信息 | Upper zone of growth plate and cartilage matrix associated (UCMA), also known as Gla-rich protein (GRP), is a highly charged, chondrocyte-specific secreted protein expressed primarily in the growth plate and articular cartilage. It undergoes cleavage into a C-terminal fragment that functions as a potent inhibitor of ADAMTS aggrecanases, such as ADAMTS5. By physically interacting with these enzymes, UCMA blocks the cleavage of aggrecan and prevents the formation of NITEGE neoepitopes induced by pro-inflammatory cytokines like IL-1β or TNFα. This mechanism protects against proteoglycan loss and cartilage degeneration. Clinically, UCMA is significantly involved in the pathogenesis of osteoarthritis and inflammatory arthritis, where it plays a dual role by reducing cartilage damage while simultaneously promoting osteophyte formation through the regulation of hypertrophic chondrocytes, osteoblasts, and osteoclasts. |
