| 背景信息 | Progesterone immunomodulatory binding factor 1 (PIBF1, also known as CEP90/C13orf24) is a multifunctional protein with distinct intracellular and secreted roles. The intracellular isoform localizes to the pericentriolar material, where it is essential for centriole duplication, mitotic spindle integrity, and ciliogenesis by recruiting key centrosomal proteins. Conversely, the secreted isoform acts as a critical mediator of progesterone signaling during pregnancy, modulating arachidonic acid metabolism and shifting immunity toward a Th2 profile via JAK/STAT signaling to prevent fetal rejection. Biallelic loss-of-function variants in PIBF1 are associated with autosomal recessive ciliopathies, including Joubert syndrome 33, highlighting its importance in developmental pathways. |
