| 背景信息 | Rho GTPase activating protein 45 (ARHGAP45), also known as HMHA1, is a negative regulator of Rho-type GTPases, specifically targeting RhoA. It utilizes its C-terminal RhoGAP domain to inactivate GTP-bound RhoA, thereby controlling actin cytoskeleton dynamics, cell spreading, and endothelial barrier function. The protein also contains an N-terminal BAR domain that provides autoinhibition. ARHGAP45 is involved in the chemokine-driven migration of hematopoietic progenitors and naive T and B cells. Clinically, ARHGAP45 is a lineage-specific dependency in acute myeloid leukemia, where its suppression of RhoA promotes leukemia cell survival. Furthermore, it is the precursor to the minor histocompatibility antigen HA-1. Mismatching of this antigen between donor and recipient in bone marrow transplantation is a primary trigger for graft-versus-host disease, making it a significant target in transplant immunology and leukemia immunotherapy. |
