| 背景信息 | ATP binding cassette subfamily C member 2 (MRP2) is an ATP-dependent efflux transporter that mediates the export of a wide range of organic anions, including conjugated bilirubin, bile salts, and various drug conjugates. It is predominantly localized to the apical membrane of hepatocytes, as well as the renal proximal tubule and intestinal epithelial cells, where it facilitates hepatic detoxification and biliary excretion. Structurally, MRP2 consists of two transmembrane domains, two nucleotide-binding domains, and a regulatory domain that modulates substrate selectivity and transport activity. This transporter plays a critical role in chemoprotection and drug disposition. Pathogenic variants in the ABCC2 gene cause Dubin-Johnson syndrome, a condition characterized by conjugated hyperbilirubinemia. Additionally, alterations in MRP2 expression or function are associated with multidrug resistance in various cancers. |
