| 背景信息 | Thymic stromal lymphopoietin (TSLP) is a cytokine primarily produced by epithelial cells and a member of the IL-7 family. It signals through a high-affinity heterodimeric receptor complex composed of TSLPR and IL-7Rα, activating downstream pathways including JAK1/JAK2-STAT5, ERK, p38 MAPK, and NF-κB. TSLP acts as a master switch in allergic inflammation by polarizing dendritic cells toward Th2 responses, promoting T-cell proliferation, and stimulating the release of Th2 cytokines such as IL-4. It further modulates the activity of innate immune cells, including eosinophils, mast cells, and basophils, by upregulating adhesion molecules and prolonging cell survival. Clinically, TSLP is overexpressed in allergic disorders such as asthma, atopic dermatitis, and allergic rhinitis, where it sustains eosinophilic infiltration and Th2-skewed immunity. |
