| 背景信息 | Autophagy related 4C cysteine peptidase (ATG4C) is a cysteine-type peptidase essential for the regulation of the autophagy pathway. It mediates the proteolytic activation and delipidation of ATG8 family proteins, including MAP1LC3 and GABARAPL2, by exposing a C-terminal glycine residue. This modification is required for the conjugation of these proteins to phosphatidylethanolamine on autophagosomal membranes and their subsequent recycling. While ATG4C exhibits weaker cleavage activity than its paralog ATG4B, it demonstrates high delipidation efficiency, contributing to the maturation and dynamism of autophagosomes. It is also involved in selective autophagy pathways and may undergo caspase-mediated activation via a specific DEVD site. Unlike other family members, ATG4C is not essential for basal autophagy, suggesting a specialized role in stress-induced or tissue-specific autophagic processes. |
