| 背景信息 | Mitochondrial transcription termination factor 3 (MTERF3) is a nuclear-encoded protein that localizes to the mitochondrial matrix, where it acts as a negative regulator of mitochondrial DNA (mtDNA) transcription initiation. By binding to mitochondrial promoter DNA, MTERF3 coordinates the balance between transcription, translation, and respiratory chain biogenesis. Structurally, it is a helical repeat protein that also interacts with mitochondrial 16S rRNA to promote the assembly of the large 39S mitochondrial ribosomal subunit. This dual role ensures efficient mitochondrial translation and the proper formation of respiratory complexes. MTERF3 serves as a critical checkpoint in mitochondrial gene expression; its loss leads to the overactivation of mtDNA transcription and defective ribosome biogenesis, ultimately resulting in impaired oxidative phosphorylation and significant mitochondrial dysfunction. |
