| 背景信息 | Vacuole Membrane Protein 1 (VMP1), encoded by the VMP1 gene on human chromosome 17 (previously TMEM49), is a transmembrane endoplasmic reticulum (ER) protein that functions as a phospholipid scramblase, facilitating lipid equilibration across ER membranes for processes like autophagosome biogenesis, where it interacts with ATG2 to support autophagy initiation even under nutrient-rich conditions. It regulates ER contacts with organelles such as lipid droplets, mitochondria, endosomes, and the Golgi, modulates ATP2A2 activity, and is essential for cell junction formation, Golgi organization, and exocytosis; subcellularly, it shows mainly cytoplasmic expression across tissues. VMP1 is overexpressed in acute pancreatitis, mediating zymophagy to degrade activated zymogen granules in acinar cells, and associates with chronic pancreatitis and pancreatic ductal adenocarcinoma via autophagy pathways; it also promotes cytoplasmic vacuolization during early pancreatitis and acts as a host factor for flavivirus infections like Zika by enabling ER remodeling for replication organelles. |
