| 背景信息 | PDZ and LIM domain 5 (PDLIM5) is a cytoplasmic adaptor and scaffold protein belonging to the PDZ-LIM family. It features an N-terminal PDZ domain that mediates protein-protein interactions with the cytoskeleton, such as alpha-actinin, and three C-terminal LIM domains that interact with kinases and transcription factors, including PKCβ, ILK, and CREB. Primarily localized to the Z-disk in striated muscles, PDLIM5 facilitates actin dynamics, mechanotransduction via the integrin-YAP pathway, and cardiomyocyte expansion. Alternative splicing produces several isoforms critical for neuronal dendritic spine morphogenesis and osteogenic maturation. In clinical contexts, dysregulation of PDLIM5 is linked to cancer progression; it enhances Ras-ERK signaling in thyroid carcinoma and stabilizes SMAD3 to promote epithelial-mesenchymal transition and metastasis in lung cancer. |
