| 背景信息 | Oligodendrocytic myelin paranodal and inner loop protein (OPALIN) is a central nervous system-specific type I transmembrane protein localized to the paranodal loops of myelinated axons and oligodendrocyte soma. It functions as a primary receptor for LGI1, utilizing specific N-terminal residues to facilitate high-affinity binding. The LGI1/OPALIN signaling axis is essential for orchestrating oligodendrocyte differentiation and terminal myelination by regulating the activity of transcription factors Sox10 and Olig2. This interaction promotes myelin gene expression and maintains white matter integrity. Clinically, OPALIN is associated with demyelinating conditions such as Multiple Sclerosis and LGI1-related disorders. Mutations or deficiencies in OPALIN result in hypomyelination and white matter abnormalities, making the LGI1/OPALIN axis a significant target for therapeutic intervention in neurodegenerative and demyelinating diseases. |
