| 背景信息 | Ring finger protein 2 (RNF2), also known as RING2, is an E3 ubiquitin-protein ligase encoded by the RNF2 gene on human chromosome 1, featuring a conserved RING finger domain that binds zinc atoms to facilitate ubiquitin conjugation. As a core component of the Polycomb repressive complex 1 (PRC1), RNF2 primarily monoubiquitinates histone H2A at lysine 119 (H2AK119Ub), imposing an epigenetic mark for transcriptional repression of developmental genes like Hox clusters, while its activity is enhanced by BMI1 and modulated by RING1. RNF2 exhibits chromatin binding and ubiquitin-protein transferase activity, associates cell-cycle dependently with DNA, interacts with proteins like TFCP2 and HIP2, and negatively regulates autophagy via AMBRA1 ubiquitination. In diseases, RNF2 links to neurodevelopmental disorders such as Luo-Schoch-Yamamoto Syndrome and non-specific syndromic intellectual disability from rare de novo missense variants in its RING domain, and overexpression promotes cancers like colorectal cancer by repressing EGR1, correlating with poor prognosis. |
