| 背景信息 | BRCA1 associated RING domain 1 (BARD1) is a critical scaffold and enzymatic partner that heterodimerizes with BRCA1 to form a functional E3 ubiquitin ligase complex. This complex mediates 'Lys-6'-linked polyubiquitination of targets such as RNA polymerase II, playing a central role in the DNA damage response. BARD1 contains an N-terminal RING finger domain for BRCA1 binding, ankyrin repeats, and tandem BRCT domains that facilitate interactions with CstF-50 for mRNA processing and RAD51 for homologous recombination repair. It also contributes to chromosomal stability, p53 stabilization, and the induction of apoptosis. Clinically, germline and somatic mutations in BARD1 are well-established risk factors for familial and sporadic breast, ovarian, and uterine cancers. These mutations typically disrupt the BARD1-BRCA1 interaction or the protein's structural integrity, thereby impairing its tumor-suppressive functions. |
