| 背景信息 | Neurturin (NRTN) is a secreted neurotrophic factor belonging to the glial cell line-derived neurotrophic factor (GDNF) family and the TGF-beta superfamily. It signals by preferentially binding to the GFRα2 co-receptor, which subsequently activates the RET receptor tyrosine kinase. This interaction triggers downstream MAP kinase and PI3K/AKT/mTOR pathways, promoting the survival, differentiation, and synaptic development of various neuronal populations, including midbrain dopaminergic and enteric neurons. Neurturin also influences non-neuronal systems such as hair follicles and immune cells. Clinically, mutations in the NRTN gene are associated with Hirschsprung disease, characterized by the absence of enteric ganglia. Additionally, elevated neurturin levels are linked to increased invasiveness in pancreatic cancer and the progression of nephroblastoma. |
