| 背景信息 | VRK serine/threonine kinase 1 (VRK1) is a nuclear kinase that regulates essential cellular processes, including cell growth, division, and chromatin organization. It phosphorylates several key targets, such as p53 to stabilize it for DNA repair, histone H3 at Thr3 to facilitate chromosome condensation during G2/M progression, and CREB to drive cyclin D1 expression. VRK1 also plays a role in nuclear envelope assembly and the organization of Cajal bodies for RNP complex assembly. Clinically, mutations in VRK1 are well-established causes of pontocerebellar hypoplasia type 1A (PCH1A), a severe neurodevelopmental disorder characterized by brain underdevelopment and microcephaly. Additionally, VRK1 variants are linked to hereditary motor sensory neuropathy, distal hereditary motor neuropathies, and spinal muscular atrophy, underscoring its critical role in both general cell division and specialized neuronal maintenance. |
