| 背景信息 | Growth Differentiation Factor 15 (GDF15) is a stress-responsive cytokine belonging to the transforming growth factor-beta (TGF-β) superfamily. Synthesized as a 308-amino acid proprotein, it undergoes proteolytic cleavage to release a disulfide-linked homodimeric mature peptide (~25 kDa). GDF15 is widely expressed in tissues under pathological stress, including inflammation, hypoxia, or mitochondrial dysfunction. It signals through a receptor complex involving GDNF family receptor α-like (GFRAL) and RET tyrosine kinase, primarily in the brainstem, regulating energy homeostasis, appetite, and body weight. Elevated GDF15 levels are linked to diverse diseases, serving as a biomarker for conditions like cancer cachexia, cardiovascular diseases (e.g., heart failure, atherosclerosis), chronic kidney disease, and obesity-related metabolic disorders. In cancer, GDF15 promotes cachexia by suppressing appetite and inducing muscle wasting, while its role in obesity involves counteracting hyperphagia. |
