| 背景信息 | 5'-nucleotidase, cytosolic IA (NT5C1A) is a cytosolic enzyme that catalyzes the hydrolysis of ribonucleotide and deoxyribonucleotide monophosphates, such as AMP, IMP, and dCMP, into nucleosides and inorganic phosphate. It is highly expressed in skeletal muscle and plays a vital role in nucleotide metabolism and the regulation of AMP/ATP ratios, which influences AMPK activation and subsequent glucose uptake. Clinically, NT5C1A is a major autoantigen in inflammatory myopathies, particularly sporadic inclusion body myositis. Anti-NT5C1A seropositivity in these patients is a specific marker that predicts severe motor, bulbar, and respiratory involvement. Additionally, NT5C1A is overexpressed in pancreatic cancer, where it contributes to gemcitabine resistance by depleting the intracellular metabolites required for the drug's efficacy, and it modulates nucleoside analog responses in various leukemias. |
